1、重组水通道蛋白9对非酒精性脂肪肝病细胞模型的作用王川,袁媛,姜政,王丕龙(400016重庆,重庆医科大学附属第一医院消化内科)摘要I目的 构建人水通道蛋白9(叫UaglyCerOPorin9,AQP9)重组质粒,验证其在L-02肝细胞中的表达,并检测其对非酒 精性脂肪肝病(nonalcoholic fally liver disease, NAFLD)细胞模型的作用。方法 从人肝脏组织中提取总RNA, RT-PCR获得AQP9 基因,克隆到载体PEGFP-Nl上,构建重组质粒PEGFP-NI-AQp9。将其转染L-02细胞,通过荧光显微镜观察其转染情况,RT-PCR 和WeStembIot检测
2、AQP9基因在细胞中的表达。通过油红O染色,测定甘油二酯、游离脂肪酸及甘油含量检测其对L-02细胞脂肪变 性模型的作用C结果成功构建PEGFP-Nl-AQP9重组质粒,并能在L-02细胞中表达,将其转染L-02细胞脂肪变性模型后,油红O 染色可见油酸pEGFP-N1-AQP9转染组较油酸组细胞内脂质含量明显升高,油酸pEGFP-NI-AQP9转染组细胞内甘油三酯、游离脂 肪酸及甘油含量分别为5.4350.337 mmol/L、2.0160.144 mmol/L&l.4850.113 nmolL,而油酸组分别为3.2180.220 mmol/L、 L5380.193mmolL及1.024 0.1
